Once Amy Coney Barrett is confirmed, should Supreme Court immediately rule that all mail ballots are unconstitutional and cannot be counted?

That sounds like something Trump thinks Amy Barrett is capable of doing.

Not how it works.  But that 4-4 ruling that ended up leaving a lower court ruling stand would be immediately reviewable if they want to take it..

No.   There is nothing Unconstitutional about mail ballots.    Even Trump uses one.

My entire state has been mail-in voting only for more than a decade and no one in either political party has said "boo" about voter fraud.

By the way, the Supreme Court doesn't make laws.

I don't like the mail in voting either because it's vulnerable to fraud, but the toothpaste is out of the tube and we're stuck with the mess.

Oh sweetie, it doesn't work like that. Try and pay a little more attention in civics next time.
