Answering the first question only as per our guidelines.
As you should know, the mRNA codes for a specific polypeptide chain with the help of codons. These codons are combination of three nucleotides whose sequence determine the type of amino acid to be added into the chain. The repetition of these codons in a specific fashion is called the reading frame of that mRNA.
The first codon that specifies the forward reading frame is usually AUG that is sequence of adenine-uracil-guanine. This codon usually codes for amino acid metheonine. This is the reason why almost all proteins have metheonine as the first amino acid in the chain.
