Is informing your employer your last day of availability to work is in two weeks different than informing them that in two weeks you’ll quit?

Same thing. You are telling them in two weeks, you will no longer be working for them.

The former is the classic “two weeks notice”. The latter is telling them your intention (may or may not actually happen) of quitting, without notice, in two weeks.
