A. After school most of them played some kind of sport.
B. After school, most of them played some kind of sport.
Do I need the comma after school?

The comma is preferred but not required. Read the sentence aloud and notice how you naturally pause between "school" and "most." In an informal context (where the comma does not influence the meaning, the coma is optional.) In a formal context (where the comma helps clarify the meaning) the comma is necessary. A better (simpler) sentence would be: Most of them played sports after school.
