We honestly have no way of knowing, since the period predates writing by about 700 years, but there was probably a number of spoken languages, including proto sumerian, as Mesopotamian society at that time appears to have been comprised of a number of disparate tribes rather than a single homogeneous unit.

Unknown. The Sumerian language is a language isolate and the oldest written language in existence. Sumer was first settled between 4500 and 4000 bce by a non-Semitic people who did not speak the Sumerian language. These people now are called proto-Euphrateans or Ubaidians.
