Invest in a travel guide book to New York with tourist information. Get an up-to-date edition. It should include public transport options.

Most of the major hotels, especially in midtown, will be able to advise you on how to get to whatever sites you wish to see. Manhattan may be regarded as divided into east and west sides by Fifth Avenue. The A, 1, 2, and 3 subways run on the west side; 4, 5, and 6 on the east.
Not all worthy locations are in Manhattan, which comprises only 21% of the land surface of NYC. Each of the 5 boroughs, for example, has a zoo your children might enjoy as each specializes.

Get a guide book, check out YouTube and there are plenty of web sites that will help you see and do what you want. You're an adult, act like one - do your research

The subway line you need depends on exactly where you're starting from, and where you want to end up. Get a map, or download a navigation app (CityMapper for example)

You have to figure out is whether you need to go uptown or downtown. As some subway stations have separate entrances for uptown or downtown-bound trains which means that you might have to cross the street to get down to the direction you want to go.
As you are traveling with kids we suggest you take Hop on Hop off buses. These are really useful for getting an overview of the city.
You can book your tour here https://freetoursbyfoot.com/new-york-tours/book-on...
Most express stations will have entry to both platforms accessible from every entrance to the station.

These are some of the subway lines in NYC:
And I probably forgot some.
There is no single subway line that covers all landmarks.
Some of them go from all the way downtown to all the way uptown. Some go to from midtown to the Bronx. Some travel only on the east side of Manhattan to the Bronx or Brooklyn, others only on the west side. Some go on the diagonal. Others go from midtown to Queens or Brooklyn. Others travel from Brooklyn to Queens.
The questions are: WHERE IS YOUR HOTEL AND WHAT TOURIST PLACES DO YOU WANT TO SEE?
So, as you can see, you have to define your terms before anyone here can even begin to help you.

There are many different subway lines. It is best that you take a look at (and download to your cellphone) the online subway map: https://new.mta.info/sites/default/files/2019-04/s...
Paper copies are free at any subway booth.
You will have to plan out what line/route to get to where you want to go.
The best hotels are in midtown but they are also the most expensive.
Without more specific information there is not much more else that can be provided.
Try https://www.nycgo.com/ for more information on hotels, retaurants etc

You need to do your own research, starting with a hotel that is centrally located, and that you can afford. You also need to decide how to get to NYC - we drive often, because it is less that 2 hours, and we are experienced NY drivers. Your idea of what is interesting to see is probably not the same as ours, so from where you are staying, you need to find how to get to the various places that interest you. WE are not your travel agents - so if you are clueless and lazy, get one.
