You know I never liked the word Best because so much talent out there equal in their own realm. The Everly Brothers really set the stage for the Harmonies that would be an inspiration for many to follow. The Beach Boys excelled in it and were a part of my youth, then a band that would become my all time fav. with blistering Blues. The Allman Brothers.
I'm going to add a latter band of three brothers who have never really gotten their due but oh my the talent they all possess. IMO Henry Garza is among the most talented guitarists of the last 13 years. Jojo Garza a great bass and Ringo Garza can play those drums.

Hmmm...I'm going to base my answer on which set of brothers had equal or comparative input into the music. On those terms I have to narrow it down between the Allmans (Gregg's songwriting + Duane's guitar mastery) and the Youngs (joint songwriters, Malcolm's rhythm work propelling the songs while Angus' lead work lights them up). I'll go with the Allmans, but it's a close pick -- they simply have the advantage of greater diversity.
Ron and Russell Mael (Sparks)
Dimebag Darrell and Vinnie Paul (Pantera)

Barry, Robin, Maurice and Andy Gibb
Jackie, Tito, Jermaine, Marlon and Michael Jackson
But I'll go with Duane and Gregg.
Edit: do the brothers have to perform together? Cuz if so, Andy wasn't a Bee Gee. Also, if they don't you'd want to consider Ronnie, Donnie and maybe Johnny Van Zant, also Johnny and Edgar Winter.
