Would this word in the sentence be plural or singular?

Plural, because you are referring to "one of" a group of populations. If you were talking about THE largest population, it would be singular, but you're talking about one out of a group of more than one. It wouldn't make sense to say "one of the largest one population."

Plural. There are many colleges etc., each with a population, small or large. Of these populations, "I see...one of the largest (of the student body populations).

Try,'When I see..., I see one of the largest populations, with over 50,000 students.'
Probably not a good idea to have the word student mentioned twice in that sentence.

After "one of the..." you need a plural word.
I see one of the books.
I ate one of the bananas.
I see one of the populations.
