1. You have to write 50 words, including punctuation.
可以。但是我建議使用 punctuation marks.
punctuation 泛指所有標點，不需加 s.
例子：for words including punctuation and hyphen characters(參考這裏，標題部份)
但我要說明的是 if...or (是否)的來由

Is this good or rotten?
I don't know if this is good or rotten.

Nothing wrong with your sentences, except, "punctuation" needs plural as "punctuations".
