How can I type this word in Japanese?
How to type kinō in Hiragana?
Is it きんう or きのう ?
AND Do I need to add う follow by all the vowel, ā, ē, ī, ō, ū? Like for Depāto is でぱうと？

It's きのう. And you should type kinou in roma-ji and its pronunciation is ki-no-u.
You should pronounce ā, ē, ī, ō, ū as aa, ee, ii, oo, uu.
Usually these are called "prolonged sound" and represented as "ー" in Japnese. So when you say Depāto, you should pronounce "Depaato (でぱあと)" and write "でぱーと". Well, what I wanna let you know is, it does not sound Dep-aa-to but Depa-a-to.

きのう is correct.
Many cases, long vowel of o needs う in a hiragana word.
Some are お like とお (ten), とおり (street), こおり (ice), or おおきい (big).
Basically long vowel of e needs い in a hiragana word.
Expecption is え in おねえさん (older sister).
All the long vowel of i needs い in a hiragana word.
All the long vowel of a needs あ in a hiragana word.
Always ー is used in a katakana word like デパート.
By the way how can I type ō?
