If you tell people to "make it count", you are telling them to make what they are doing as useful and as positive as possible. They should not be half-hearted but give the task their whole attention and enthusiasm. In the sentence "Live life to the full and make every day count", this means "Don't waste a single day by being being lazy or useless - do something positive every day".

I saw that film a long time ago but if you are talking about then end then I took it to mean that he wanted her to live her life to it's fullest so that his sacrifice was worth while - afterall who wants to sacrifice themself for someone who then goes on to live an un fulfilled life as a petty criminal!?
