Materials and methods

2.1. Plant material

Salvia miltiorrhiza flower was collected at seven locations in China

in 2006 (Table 1). All samples were air-dried at room temperature

in a shady place and kept from direct light.

Hydrodistillation was used for 6 h to extract the essential oils.

Distillate was extracted with ether, and then frozen at 18 C to remove


2.2. GC analysis

Analytical gas chromatography was carried out using a Shimadzu

(Osaka, Japan) GC-2010A series gas chromatograph with an

OV-1701 capillary column (30 m 0.32 mm, 0.33 lm film thickness).

Nitrogen was the carrier gas. Column temperature was initially

kept at 70 C for 5 min, then gradually increased to 280 C

at a rate of 5 C/min, and held for 10 min. Samples of 1 ll were injected

at 280 C, manually with split mode. Flame ionisation detection

(FID) was set at 280 C.

2.3. GC–MS analysis

GC–MS analysis was performed on a 6890N gas chromatograph

with fused silica capillary column (J&W HP-5, 30 m 0.25 mm,

0.25 lm film thickness), coupled to a mass selective detector

(MSD5973N, ionisation voltage 70 eV; all Agilent, Santa Clara,

CA). Helium was used as carrier gas. Oven temperature was from

80 C to 290 C at 4 C/min and held at 290 C for 30 min. The components

were identified by comparison of their mass spectra with

those of the NIST02L mass spectral library.

    氣相層析儀分析儀器主要用三津(大陸翻譯為島津)日本大阪(Osaka, Japan)型號為GC-2010A系列並用OV-1701的毛細管柱(30米,0.32mm口徑,0.33um膜厚)。載流氣體為氮氣。管柱條件初始溫度為70℃維持5分鐘,然後以每分鐘升5℃到280℃並維持10分鐘。樣品注射體積為1ul注射口溫度280℃,手動設分流模式。火焰離子偵測器溫度設定為280℃。

    2.3. 氣相層析質譜儀分析

    氣相層析質譜儀主要用6890N氣相層析儀執行;並用玻璃纖維的毛細管柱(J&W廠牌 HP-5型號,30 米 0.25 mm口徑,0.25um 膜厚),連接質譜儀( 質譜儀5973N型,離子化電壓70eV;均為安捷倫廠牌,美國加州聖克拉拉),氦氣作為載氣,烘箱溫度從80℃以每分鐘升4℃至290℃並維持30分鐘。樣品組成份鑑定則是利用NIST02L質譜圖庫作為判別比對樣品質譜。
