The present study showed that oxytocin increased proliferation of
SaOS-2 cells whereas it inhibited proliferation of TE-85 cells. In spite
of these opposite effects of oxytocin, the most effective
concentrations of oxytocin were similar in the two types of cells, and
the effects were abolished by an oxytocin antagonist. The strongest
effect of oxytocinwas induced when the cells were incubated with
oxytocin during 24 h, which is reasonable since the half-life of
oxytocin is very short (minutes). In addition, oxytocin inhibited cell
proliferation of a rat osteosarcoma cell line, UMR-106.
Thymidine incorporation measures DNA synthesis and is a
wellestablished method for indirect measurements of cell proliferation.
However, it has one disadvantage since also cells which not are viable
at the time of measurement are registered. Therefore we used the
commercially available kit EZ4U as a complement. This kit is based on
the capability of living cells to reduce slightly coloured or uncoloured
tetrazolium salts into intensely coloured formazon derivates. The two
methods gave almost similar results, except that also lower
concentrations of oxytocinwere efficient when analyzed by the
EZ4U-kit. The reason behind this discrepancy might be that the
experimental groups (number of wells) in the experiments analyzed
by the EZ4U kit were larger and thereby easier could reach statistical
We have in previous studies found that oxytocin stimulates
proliferation of human osteoblast-like cells and SaOS-2 cells. In
contrast, oxytocin has also been reported to inhibit proliferation of
some other cell lines including some osteosarcoma cell lines.
Therefore we investigated the effects of oxytocin on cell proliferation
in two human osteosarcoma cell lines known to represent different
stages of osteoblast differentiation. The human osteosarcoma cell line
SaOS-2 is rather highly differentiated and expresses both osteocalcin
and ALP which are well-known osteoblastic markers.
