Thiamine diphosphate dependent lyases

Well-known examples of thiamine diphosphate dependent

lyases are pyruvate decarboxylase (PDC; EC

4.1.1.1), benzoylformate decarboxylase (EC 4.1.1.7),

and benzaldehyde lyase. Pohl et al. [14] have reviewed

the characteristics of this group of biocatalysts extensively,

while Ward and Singh [15] summarise the use

of decarboxylases for asymmetric synthesis. The industrial

utilization of PDC in the chemoenzymatic synthesis

of ephedrine was established in the 1930s. Yeast producing

high amounts of PDC is used to synthesise Rphenylacetylcarbinol

from pyruvate and benzaldehyde;

R-phenylacetylcarbinol is then chemically converted to

ephedrine. Unlike the yeast biocatalyst, PDC from Zymomonas

mobilis also accepts considerable amounts of acetaldehyde

instead of pyruvate as substrate for the C–C

coupling reaction. The enzyme from Zymomonas was

optimised by protein engineering to increase its aldehyde

stability [16]. Recent progress with PDC was also made

using two-phase systems for the biocatalytic synthesis of

R-phenylacetylcarbinol [17,18].
