depending on what os you are running you might want to check your firewall settings. if its windows xp check the windows firewall to make sure that connection protocols initiated by itunes are allowed and not blocked. you can check this by making sure that itunes progam is allowed to access the outside network. if still same try restarting the computer or reinstalling itunes as a last alternative.
