請問有人知道JunB跟造血幹細胞之間的關係嗎???

是有一篇PAPER是有關造血幹細胞的

跟裡面有很多機制~以我們現在所學的~完全無法理解~

希望有人看過這篇PAPER的可以跟我們說明一下~

是CELL這的雜誌~

    根據內文,他說 對長期性造血幹細胞 (LT-HSC)而言,JunB是一個主要的負調控因子。

    因為JunB不表現的時候,只有幹細胞時期比較容易發展成骨髓增生性疾病(myeloproliferative disorder, MPD),而唯一跟JunB缺陷有關的幹細胞就是LT-HSC(包括白血球幹細胞, leukemic stem cells -LSC)。

    那JunB其實是調控 LT-HSC的數量,因為他是一個transcription factor嘛。可能就是透過對幾個主要基因的表現調節去影響幾個機制,像是增殖proliferation,老化senescence和細胞自殺apoptosis等等。

    那這些被JunB調控的些基因在文章裡提到的比如說像是p16INK4a,他是一個cell cycle regulator。Bcl2 and Bclxl的話就是抑制apoptosis的蛋白。

    綜合以上內容,就是說當 JunB壞掉的時候,將會無法調節 LT-HSC的數量而導致大量增生的情形,而造成骨髓增生性疾病 MPD。

    JunB as a key negative regulator of LT-HSC function, controlling the numbers of LT-HSC by regulating several cellular mechanisms that are critical for maintenance of the stem cell compartment, i.e., proliferation/senescence and apoptosis, via changes in the expression levels of crucial effector genes, including the cell cycle regulators p16INK4a and the antiapoptotic proteins Bcl2 and Bclxl.

    玩這麼難的

    臺灣現在水準這麼高

    這paper難

    難在 transgenic mouse breeding

    而不在junB 的mechanism

    一JunB 是transcriptional regulator 屬於AP-1 transcriptional factor family

    二目前已知 JunB 可以促進一個叫p16ink4a cell cycle inhibitor 蛋白的表現

    減低兩個antiapoptotic 蛋白 Bcl2 and Bclxl的表現

    三inactivation of JunB 則會造成老鼠出生後的 myeloproliferative disorder (MPD)

    這和人的 chronic myelogenous leukemia (CML)很像

    四overexpression of JunB 則在造血幹細胞的分化上結果減少long-term-

    hematopoietic stem cells(HSC) 數目,反之, inactivation of JunB 則使得long-term- hematopoietic stem cells(LT-HSC) and Graunocytes/ marcrophage progtenitor數目增加並造成老鼠的myeloproliferative disorder (MPD)

    五將帶有inactive JunB 的long-term- hematopoietic stem cells(LT-HSC)做

    骨髓移植到特定的transgenic老鼠身上則會造成老鼠的myeloproliferative disorder

    六將 enforced expression JunB的long-term- hematopoietic stem cells(LT-HSC)

    做骨髓移植到特定的transgenic老鼠上, 這個population 的stem cell 會消失, 這是 JunB 蛋白作

    用在促進p16ink4a cell cycle inhibitor 蛋白的表現和降低antiapoptotic

    蛋白 Bcl2 and Bclxl的表現的結果

    七這些結果證明了JunB在stem cell中扮演著特別的角色並且stem cell在分化在

    long-term- hematopoietic stem cells(LT-HSC) 階段時,細胞可以做骨髓移植使用,

    並構造一個老鼠myeloproliferative disorder (MPD) 的動物model

    A:在造血細胞的分化過程中JunB蛋白主要表達在LT-HSC, GMP和Granulocytes

    這三個階段的細胞中, inactivation of JunB會造成clonal expansion of LT-HSC,

    GMP and Granulocytes,這是肇因於 increased proliferation and blockade

    What is function of JunB the author did not give any advanced articulation at all. All the information on JunB was claimed by former researchers. This paper can be published in “Cell”. I am surprised. Perhaps, it is normal. On earth, the impact factor for cell is escalating. I guess its impact factor is getting low and low. In this year, a editor in edictal broad did the “paper retraction". In cell biology sphere, some people are going to discuss why this hasppened. Of cause, those criticisms are negative to journal “cell” and perhaps might cause negative effect.
