What advances in technology would be required to send a manned mission to Mars?

Most sources seem to be predicting that we will send astronauts to land on Mars in about 25-50 years, what is the difference between sending the mission in 2011 and 2045, what will be able to do better in 2045 or 2075 that we cannot do now?

we would need the ability to generate artificial gravity otherwise the astronauts muscles would waste away, we would need to have line of communication satellites near to mars and earth to relay communications, a new form a propulsion would be needed as conventional rocket fuel is expensive and a large amount would be needed, something like a ion propulsion System would be good. we would also need a way to recycle every thing that they use on board the ship, so water even used toilet water, food and oxygen. more advanced shielding to protect the ship from extreme forms of radiation or solar flares which may occur. there are many more advances that we would need, so i would say another 25 - 30 years before we go any where near mars.
